The alleged illegitimacy of Edward V1 would not affect Scotland. However there is an alternative Royal line in Scotland, in direct descent from Charles 2nd which could lay claim to the throne.
In 1663, Anne Scott, Countess of Buccleuch in her own right, married James 'Crofts' the eldest son of Charles 2nd. (The name Crofts was the name of his adoptive father.)
James took the name Scott on his marriage to the Countess and was created Duke of Monmouth and Buccleuch. He was alleged to be illegitimate, having been born while Charles, then Prince of Wales was still in exile in Holland. At that time, restoration of the Monarchy seemed unlikely and Charles married Lucy Walters.
Charles's mother, Queen Henrietta Marie was furious when she heard of the marriage and threatened to have her brother, the King of France, cut off the substantial pension which Chares received from him if Charles did not repudiate Lucy and her son James.
The prince was forced to do so and later married Catherine of Braganza in 1662, four years after the death of Lucy Walters.
Charles died in 1685 without any acknowledged legitimate heirs and was succeeded by his brother, James, Duke of York.
Monmouth, the true heir, rebelled but was defeated and executed, however, through his marriage to the Countess of Buccleuch, he left several children. His eldest son, the Earl of Doncaster died young but the second son, the Earl of Dalkeith produced a large family and his eldest son succeeded his grandmother as Duke of Buccleuch.
The direct descendent of the Duke of Monmouth is the current Duke of Buccleuch and Queensberry, and, as a direct descendent of King Charles 2nd, he is the rightful heir of the Royal House of Stewart, it's titles, and all that this implies.
